Comments

Phenol will take up approx. 25 % water, if water-saturated. This phenol only contains approx. 9 % of water. Therefore, a lot of water will be taken up from the aqueous, RNA-containing sample during RNA extraction. The advantage of the water-uptake is a reduction of the sample volume (concentration of the RNA). It might be a disadvantage, if the sample already contains high salt concentrations, because salt will be concentrated too.
